N/OFQ (IHC-grade), Nociceptin/Orphanin FQ Antibody

Figure 1. Immunohistochemical identification of Nociceptin/Orphanin FQ in rat dorsal horn. Sections were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, Nociceptin/Orphanin FQ was detected in a dense plexus of fibres and terminals in the superficial dorsal horn.

Figure 2. Immunocytochemical analysis of Nociceptin/Orphanin FQ expression in N/OFQ+/+ and N/OFQ-/- mouse brains. Coronal sections of wildtype (N/OFQ +/+) and Nociceptin/Orphanin FQ knockout (N/OFQ/-/-) mice were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, Nociceptin/Orphanin FQ staining was only detected in wildtype mice in mice in the ventromedial hypothalamic nucleus, amygdala, and habenular nucleus.

Immunofluorescent identification of Nociceptin/Orphanin FQ in rat spinal trigeminal nucleus. Sections were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100 or an mouse anti-Enkephalin antibody. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Color was developed by incubation with streptavidine-cyanine 2.18 (green) and anti-mouse-cyanine 5.18 (red). Note, Nociceptin/Orphanin FQ  (green) and Enkephalin (red) were distributed in the same tissue but show no co-localization.
