EP4 receptors preferentially couple through Gs protein to stimulate adenylate cyclase. The functional selectivity of natural and synthetic EP4 receptor agonists has been assessed in terms of cAMP production and β-arrestin recruitment. Currently there is interest in selective EP4 receptor antagonists as anti-inflammatory agents.EP4 receptors are regulated by phosphorylation of carboxyl-terminal serine364/serine366 (pS364/pS366-EP4), threonine369/serine370 (pT369/pS370-EP4) and serine374/serine377 (pS374/pS377-EP4). This nomenclature refers to the human EP4 receptor. This phosphorylation motif is conserved across species but corresponds to pS392/pS394-EP4, pT397/pS398-EP4 and pS402/pS405-EP4 in mice and pS367/pS369-EP4, pT371/pS372-EP4 and pS377/pS380-EP4 in rats.
