Most effects of PTH are mediated by PTH1 receptor, which is a member of Family B GPCRs. PTH1 receptor expression is particularly high in skeletal tissues and kidney but it has a nearly ubiquitous tissue distribution. PTH1 receptor signaling via Gs or Gq is regulated in a cell- and ligand-specific manner by β-arrestins and the PDZ protein NHERF1. PTH1 activity is regulated by phosphorylation of carboxyl-terminal serine493/serine495 (pS493/pS495-PTH1) and threonine503/serine504 (pT503/pS504-PTH1). This nomenclature refers to the human PTH1 receptor. This phosphorylation motif is highly conserved and is identical in mice, rats and humans. PTH1 receptor expression is particularly high in skeletal tissues and kidney but it has a nearly ubiquitous tissue distribution.
