The VPAC1 receptor, which responds to VIP and PACAP with comparable affinity. This receptor is widely distributed in the CNS, most abundantly in the cerebral cortex and hippocampus, in peripheral tissues including liver, lung and intestine and in T lymphocytes. Selective VPAC1 receptor agonists and antagonists have been reported. VPAC1 receptor desensitization, β-arrestin recruitment and internalization are regulated by phosphorylation of carboxyl-terminal serine422/serine425 (pS422/pS425-VPAC1), threonine429/serine431/threonine432 (pT429/pS431/pT432-VPAC1) and serine435/serine438 (pS435/pT438-VPAC1). This nomenclature refers to the human VPAC1 receptor. This phosphorylation motif is highly conserved across species but corresponds to pS427-VPAC1, pT431/pS433/pT434-VPAC1 and pS437/pT440-VPAC1 in mice and rats.
