Threonine429/Serine431/Threonine432 (T429/S431/T432) is a major phosphorylation site of the VPAC1 receptor. The pT429/pS431/pT432-VPAC1 antibody detects phosphorylation in response to high-efficacy agonists. T429/S431/T432 phosphorylation is a key regulator of VPAC1 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine429/Serine431/Threonine432 phosphorylation of VPAC1 Receptor. Upper panel, HEK293 cells stably expressing the VPAC1-Receptor (VPAC1) were either not exposed or exposed to 1 µM of specific VPAC1-Receptor agonist PACAP-27 for 30 minutes. Cells were lysed and immunoblotted with the anti-pT429/pS431/pT432-VPAC1 antibody (7TM0371B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-VPAC1 antibody (7TM0371N) at a dilution of 1:1000 to confirm equal loading of the gel.
