Serine422/Serine425 (S422/S425) is a major phosphorylation site of the Calcitonin receptor. The pS422/pS425-CT antibody detects phosphorylation in response to high- and low-efficacy agonists and after PKC activation. S422/S425 phosphorylation is a key regulator of CT desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the Calcitonin Receptor (CT) were either not exposed or exposed to 100 nM specific agonist CGRP for 30 minutes. Cells were lysed and immunoblotted with the anti-pS422/pS425-CT antibody (7TM0043A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-CT antibody to confirm equal loading of the gel.
