The CCK1 receptor antibody is directed against the distal end of the carboxyl-terminal tail of human Cholecystokinin Receptor 1 (previously called CCKA). It can be used to detect total CCK1 receptors in Western blots independent of phosphorylation. The CCK1 antibody can also be used to isolate and enrich CCK1 receptors from cell and tissue lysates. It also detects CCK1 in cultured cells and tissue sections by immunohistochemistry.

Figure 1. Immunohistochemical identification of Cholecystokinin Receptor 1 in gastric mucosa. Sections were dewaxed, microwaved in citric acid, and incubated with anti-CCK1 (Cholecystokinin Receptor 1) antibody (7TM0076N-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, CCK1 receptors were uniformly detected at the plasma membrane of chief cell in the gastric mucosa.

Note, CCK1 receptors were detected predominantly at the level of plasma membrane in neuroendocrine tumor.

Figure 3. Immunocytochemical identification of Cholecystokinin Receptor 1 in HEK293 cells. HEK293 cells stably expressing the Cholecystokinin Receptor 1 (CCK1) were either not exposed or exposed to 1 μM CCK-8 (Cholecystokinin-8) for 30 min and immunocytochemically stained with anti-CCK1 (Cholecystokinin Receptor 1) antibody (7TM0076N-IC) at a dilution of 1:200. Note, CCK1 receptors were confined to the plasma membrane in untreated cells (0 min). CCK1 receptors were seen in perinuclear clusters of vesicles after 30 min CCK-8 exposure.

Figure 4. Validation of the Cholecystokinin Receptor 1 in transfected HEK293 cells. Native HEK293 cells (MOCK) or HEK293 cells stably expressing the Cholecystokinin Receptor 1 (CCK1) were lysed and immunoblotted with the anti-CCK1 antibody (7TM0076N-IC) at a dilution of 1:1000.
