GPR22 (G Protein-Coupled Receptor 22) is an orphan GPCR with constitutive activity, primarily coupling to Gi/o proteins, which leads to decreased intracellular cAMP levels. Its endogenous ligand has not been identified, and its physiological functions are still being elucidated. GPR22 is mainly expressed in the heart, particularly in cardiomyocytes, as well as in the brain, including the cerebellum and hippocampus. Its expression in cardiac tissue suggests a role in cardiac function and remodeling, especially in response to stress or injury. GPR22 is under investigation as a potential pharmacological target in cardiovascular disease, particularly for its possible involvement in cardiac protection, hypertrophy, and heart failure. For more information on GPR22 pharmacology please refer to the IUPHAR database.
