The SST3 receptor antibody is directed against the distal end of the carboxyl-terminal tail of human Somatostatin Receptor 3. In can be used to detect total SST3 receptors in Western blots independent of phosphorylation. The SST3 antibody can also be used to isolate and enrich SST3 receptors from tissue lysates. It also detects SST3 in cultured cells and tissue sections by immunohistochemistry.

Figure 1. Immunohistochemical identification of Somatostatin Receptor 3 in human growth-hormone-producing pituitary adenoma tissue. Sections were dewaxed, microwaved in citric acid, and incubated with anti-SST3 (Somatostatin Receptor 3) antibody (7TM0357N-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Color was developed by incubation in 3-amino-9-ethylcarbazole (AEC), and sections were counterstained with hematoxylin. Note, SST3 receptors were uniformly detected at the plasma membrane of nearly all tumor cells.

Figure 3. Immunocytochemical identification of Somatostatin Receptor 3 in transfected HEK293 cells. HEK293 cells stably expressing the Somatostatin Receptor 3 (SST3) were either not exposed or exposed to 1 μM SRIF (somatotropin release inhibiting factor) for 30 min and immunocytochemically stained with anti-SST3 (Somatostatin Receptor 3) antibody (7TM0357N-IC) at a dilution of 1:200. Note, SST3 receptors were confined to the plasma membrane in untreated cells (0 min). SST3 receptors were seen in perinuclear clusters of vesicles after 30 min SRIF exposure.
