The NTS2 antibody is directed against the distal end of the carboxyl-terminal tail of human Neurotensin Receptor 2. It can be used to detect total NTS2 receptors in Western blots independent of phosphorylation. The NTS2 antibody can also be used to isolate and enrich NTS2 receptors from cell and tissue lysates. It also detects NTS2 in cultured cells and tissue sections by immunohistochemistry.

Figure 1. Immunohistochemical identification of Neurotensin Receptor 2 in gastric mucosa. Sections were dewaxed, microwaved in citric acid, and incubated with anti-NTS2 (Neurotensin receptor 2) antibody (7TM0310N-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, NTS2 receptors were uniformly detected at the plasma membrane of parietal cells in the gastric muscosa.
Figure 2. Immunohistochemical identification of Neurotensin Receptor 2 in exocrine pancreas. Sections were dewaxed, microwaved in citric acid, and incubated with anti-NTS2 (Neurotensin receptor 2) antibody (77TM0310N-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, NTS2 receptors were detected predominantly at the level of plasma membrane of cells in exocrine pancreas.
Figure 3. Immunocytochemical identification of Neurotensin Receptor 2 in HEK293 cells. HEK293 cells stably expressing the Neurotensin Receptor 2 (NTS2) were either not exposed or exposed to 1 μM Neurotensin for 30 min and immunocytochemically stained with anti-NTS2 (Neurotensin receptor 2) antibody (7TM0310N-IC) at a dilution of 1:200. Note, NTS2 receptors were confined to the plasma membrane in untreated cells (0 min). NTS2 receptors were seen to be translocated to cytosol after 30 min Neurotensin exposure.
Figure 4. Validation of the Neurotensin receptor 2 in transfected HEK293 cells. Native HEK293 cells (MOCK) or HEK293 cells stably expressing the Neurotensin Receptor 2 (NTS2) were lysed and immunoblotted with the anti-NTS2 antibody (7TM0310N-IC) at a dilution of 1:1000.
