Serine445 (S445) is major phosphorylation site of human GCGR receptor. The pS445-GCGR antibody detects phosphorylation in response to agonists. S445 phosphorylation is likely to be involved in efficient ligand sequestration by the Glucagon Receptor.

Figure 1. Agonist-induced Serine445 phosphorylation of Glucagon Receptor. Upper panel, HEK293 cells stably expressing the glucagon receptor (GCGR) and transiently GRK2 (Ratio GCGR receptor:GRK2 = 10:1) were preincubated with serum-free media. After 2 hours cells were either not exposed or exposed to 100 nM of Glucagon for 30 minutes. Cells were lysed and immunoblotted with the anti-pS445-GCGR antibody (7TM0251A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-GCGR antibody (7TM0251N) to confirm equal loading of the gel.
