Serine358/Threonine360/Serine361 (S358/T360/S361) are a major phosphorylation sites of the CXCR3 receptor. The pS358/pT360/pS361-CXCR3 detects phosphorylation in response to high- and low-efficacy agonists. S358/T360/S361 phosphorylation is a key regulator of CXCR3 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the CXC Chemokine Receptor 3 (CXCR3) were either not exposed or exposed to 100 nM specific CXC Chemokine receptor 3 agonist CXCL11 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS358/pT360/pS361-CXCR3 antibody (7TM0070A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent CXCR3 antibody (7TM0070N) at a dilution of 1:1000 to confirm equal loading of the gel.
