Serine434/Serine441 (S437/S441) is a major phosphorylation site of the CRF1 receptor. The pS434/pS437-CRF1 antibody detects phosphorylation in response to high-efficacy agonists. S434/S437 phosphorylation is a key regulator of CRF1 desensitization, β-arrestin recruitment and internalization. S434/S437 also undergoes constitutive phosphorylation.

Upper panel, HEK293 cells stably expressing the CRF1-Receptor (CRF1) were either not exposed or exposed to 1 µM of specific CRF-Receptor agonist CRH for 30 minutes. Cells were lysed and immunoblotted with the anti-pS434/pS437-CRF1 antibody (7TM0212C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with a phosphorylation-independent anti-CRF1 antibody to confirm equal loading of the gel.
