Serine300/Serine303 (S300/S303) is a major phosphorylation site of the human GPR35 receptor. The pS300/pS303-hGPR35 antibody detects phosphorylation in response to high-efficacy agonists. S300/S303 phosphorylation is a key regulator of human GPR35 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells transiently expressing the eYFP-tagged human GPR35-Receptor were either not exposed (vehicle) or exposed to 100 nM of specific GPR35-Receptor agonist zaprinast for 5 minutes. Cells were lysed and receptor constuct enriched with a GFP-trap before immunoblotting with the anti-pS300/pS303-hGPR35 antibody (7TM0102A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with a phosphorylation-independent anti-GFP antibody to confirm equal loading of the gel.

Flp-ln TREx 293 cells stably expressing hGPR35a-HA were either treated with vehicle or 100 μM zaprinast for 5 min and immunocytochemically stained with anti-pS300/pS303-hGPR35 antibody (7TM0102A) at a dilution of 1:200.
