Serine323 (S323) is a major phosphorylation site of the P2Y12 receptor. The pS323-P2Y12 antibody detects phosphorylation in response to high- and low-efficacy agonists and after PKC activation. S323 phosphorylation is a key regulator of P2Y12 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the P2Y Purinoceptor 12 (P2Y12) were either not exposed or exposed to 1 µM ATP (Adenosintriphosphate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS323-P2Y12 antibody (7TM00328A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-P2Y12 antibody to confirm equal loading of the gel.
