Serine354/Threonine355 is a major phosphorylation site of the 5-HT4 receptor. The pS354/pT355-5-HT4 antibody detects phosphorylation in response to high-efficacy agonists but not after PKC activation. S354/T355 phosphorylation is a key regulator of 5-HT4 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the 5-Hydroxytryptamine Receptor 4 (5-HT4) were either not exposed or exposed to 100 µM serotonin or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS354/pT355-5-HT4 antibody (7TM0009C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-5-HT4 antibody to confirm equal loading of the gel.
