Threonine351 (T351) is a major phosphorylation site of the P2Y4 receptor. The pT351-P2Y4 antibody detects phosphorylation in response to high- and low-efficacy agonists and after PKC activation. T351 phosphorylation is a key regulator of P2Y4 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the P2Y Receptor 4 (P2Y4) were either not exposed or exposed to 1 µM ATP (Adenosintriphosphate) or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pT351-P2Y4 antibody (7TM00325C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-P2Y4 antibody to confirm equal loading of the gel.
