Serine364/Threonine365 (S364/T365) is a major phosphorylation site of the TRH1 receptor. The pS364/pT365-TRH1 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. S364/T365 phosphorylation is primarily mediated by GRKs and is a key regulator of TRH1 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the TRH1-Receptor (TRH1) were either not exposed or exposed to 1 µM of specific TRH1 agonist Taltirelin for 30 minutes. Cells were lysed and immunoblotted with the anti-pS364/pT365-TRH1 antibody (7TM0363B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-TRH1 antibody to confirm equal loading of the gel.
