The N/OFQ receptor antibody is directed against human Nociceptin/Orphanin FQ. The N/OFQ antibody can be used to detect N/OFQ in formalin-fixed, paraffin-embedded tissue sections by immunohistochemistry.

Figure 1. Immunohistochemical identification of Nociceptin/Orphanin FQ in rat dorsal horn. Sections were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, Nociceptin/Orphanin FQ was detected in a dense plexus of fibres and terminals in the superficial dorsal horn.

Coronal sections of wildtype (N/OFQ +/+) and Nociceptin/Orphanin FQ knockout (N/OFQ/-/-) mice were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100. Note, Nociceptin/Orphanin FQ staining was only detected in wildtype mice in mice in the ventromedial hypothalamic nucleus, amygdala, and habenular nucleus.

Sections were dewaxed, microwaved in citric acid, and incubated with anti-N/OFQ (Nociceptin/Orphanin FQ) antibody (7TMNOFQ-IC) at a dilution of 1:100 or an mouse anti-Enkephalin antibody. Sections were then sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Color was developed by incubation with streptavidine-cyanine 2.18 (green) and anti-mouse-cyanine 5.18 (red). Note, Nociceptin/Orphanin FQ (green) and Enkephalin (red) were distributed in the same tissue but show no co-localization.
