Serine317/threonine318 (S317/T318) is a major phosphorylation site of the D2 dopamine receptor. The pS317/pT318-D2 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. S317/T318 phosphorylation is a key regulator of D2 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Serine317/Threonine318 phosphorylation of the Dopamine Receptor 2. Upper panel, HEK293 cells stably expressing the Dopamine Receptor 2 (D2) were either not exposed or exposed to 10 μM Quinprirole or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS317/pT318-D2 antibody (7TM0215A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-D2 antibody (7TM0215N-WB) at a dilution of 1:1000 to confirm equal loading of the gel.
Figure 2. Analysis of dose-dependent Dopamine Receptor 2 phosphorylation. Upper panel, HEK293 cells stably expressing the Dopamine Receptor 2 (D2) were either not exposed or exposed to increasing concentrations of Quinpirole ranging from 1 nM to 10 μM for 30 minutes.
