Threonine355/Serine356 is a major phosphorylation site of the 5-HT6 receptor. The pT355/pS356-5-HT6 antibody detects phosphorylation in response to high-efficacy agonists but not after PKC activation. T355/S356 phosphorylation is a key regulator of 5-HT6 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the 5-Hydroxytryptamine Receptor 6 (5-HT6) were either not exposed or exposed to 100 µM serotonin for 30 minutes. Cells were lysed and immunoblotted with the anti-pT355/pS356-5-HT6 antibody (7TM0011B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-5-HT6 antibody to confirm equal loading of the gel.
