Threonine355 (T355) is a major phosphorylation site of the CCR10 receptor. The pT355-CCR10 antibody detects phosphorylation in response to high- and low-efficacy agonists and after PKC activation. T355 phosphorylation is a key regulator of CCR10 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine355 phosphorylation of the Chemokine Receptor 10. Upper panel, HEK293 cells stably expressing the Chemokine Receptor 10 (CCR10) were either not exposed or exposed to 100 nM specific Chemokine receptor 10 agonist CCL27 or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pT355-CCR10 antibody (7TM0067B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-CCR10 antibody to confirm equal loading of the gel.
