Serine336/Serine337 (S336/S337) is a major phosphorylation site of the CCR5 receptor. The pS336/pS337-CCR5 antibody detects phosphorylation in response to high- and low-efficacy agonists and after PKC activation. S336/S337 phosphorylation is a key regulator of CCR5 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Serine336/Serine337 phosphorylation of the Chemokine Receptor 5. Upper panel, HEK293 cells stably expressing the Chemokine Receptor 5 (CCR5) were either not exposed or exposed to 100 nM specific Chemokine receptor 5 agonist CCL5 or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS336/pS337-CCR5 antibody (7TM0062A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-CCR5 antibody to confirm equal loading of the gel.
