Serine324/serine325 (S324/S325) is a major phosphorylation site of the CXCR4 receptor. The pS324/pS325-CXCR4 antibody detects phosphorylation in response to agonists but not after PKC activation. S324/S325 phosphorylation is a key regulator of CXCR4 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the HA-tagged CXC Chemokine Receptor 4 were either not exposed or exposed to 100 nM CXCL12 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS324/pS325-CXCR4 antibody (7TM0071A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-HA antibody (7TM000HA) at a dilution of 1:1000 to confirm equal loading of the gel.
