Serine325/Threonine327 (S325/T327) is a major phosphorylation site of the CXCR6 receptor. The pS325/pT327-CXCR6 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. S325/T327 phosphorylation is a key regulator of CXCR6 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Serine325/Threonine327 phosphorylation of the CXC Chemokine Receptor 6. Upper panel, HEK293 cells stably expressing the CXC Chemokine Receptor 6 (CXCR6) were either not exposed or exposed to 100 nM specific CXC Chemokine receptor agonist CXCL16 or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS325/pT327-CXCR6 antibody (7TM0073B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-CXCR6 antibody to confirm equal loading of the gel.
