Serine372/Serine373 (S372/S373) is a major phosphorylation site of the D1 receptor. The pS372/pS373-D1 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. S372/S373 phosphorylation is a key regulator of D1 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine354 phosphorylation of the Dopamine Receptor 1. Upper panel, HEK293 cells stably expressing the Dopamine Receptor 1 (D1) were either not exposed or exposed to 100 µM Dopamine or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pS372/pS373-D1 antibody (7TM0214B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-D1 antibody to confirm equal loading of the gel.
