Threonine335/Serine338 (T335/S338) is a major phosphorylation site of the MCH2 receptor. The pT335/pS338-MCH2 antibody detects phosphorylation in response to high-efficacy agonists. T335/S338 phosphorylation is a key regulator of MCH2 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine335/Serine338 phosphorylation of MCH2 Receptor. Upper panel, HEK293 cells stably expressing the MCH2-Receptor (MCH2) were either not exposed or exposed to 1 µM of MCH2-Receptor agonist MCH for 30 minutes. Cells were lysed and immunoblotted with the anti-pT335/pS338-MCH2 antibody (7TM0281A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-MCH2 antibody to confirm equal loading of the gel.
