Threonine401/Serine403 (T401/S403) is a major phosphorylation site of the Orexin receptor 2 (OX2). The pT401/pS403-OX2 antibody detects phosphorylation in response to high-efficacy agonists. T401/S403 phosphorylation is a key regulator of OX2 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the Orexin Receptor 2 (OX2) were either not exposed or exposed to 100 nM Orexin-B for 30 minutes. Cells were lysed and immunoblotted with the anti-pT401/pS403-OX2 antibody (7TM0322A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-OX2 antibody to confirm equal loading of the gel.
