Serine348/Serine350 (S348/S350) is major phosphorylation site of the Atypical Chemokine Receptor 2 (ACKR2). The pS348/pS350-ACKR2 antibody detects phosphorylation in response to agonists. S348/S350 phosphorylation is likely to be involved in efficient ligand sequestration by ACKR2.

Upper left panel, HEK293 cells stably expressing the Atypical Chemokine receptor 2 (ACKR2) were either not exposed or exposed to 100 nM CCL-2 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS348/pS350-ACKR2 antibody (7TM0314A) at a dilution of 1:1000. Upper right panel, same treatment as in upper left panel but cells were additionally treated with λ-phosphatase (λ-PPase). Lower panels, blots were stripped and reprobed with the phosphorylation-independent anti-ACKR2 antibody (7TM0314N-WB) to confirm equal loading of the gel. Note that basal and agonist induced Serine348/Serine350 phosphorylation is abbolished by λ-PPase treatment.
