Threonine382/Threonine386 (T382/T386) is a major phosphorylation site of the D5 receptor. The pT382/pT386-D5 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. T382/T386 phosphorylation is a key regulator of D5 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine382/Threonine386 phosphorylation of the Dopamine Receptor 5. Upper panel, HEK293 cells stably expressing the Dopamine Receptor 5 (D5) were either not exposed or exposed to 100 µM Dopamine or 0.1 μM PMA (Phorbol 12-Myristate 13-Acetate) for 30 minutes. Cells were lysed and immunoblotted with the anti-pT382/pT386-D5 antibody (7TM0218A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-D5 antibody to confirm equal loading of the gel.
