Serine286/Threonine287/Serine288 is a major phosphorylation site of the M2 Muscarinic Acetylcholine Receptor. The pS286/pT287/pS288-M2 antibody detects phosphorylation in response to high-efficacy agonists but not after PKC activation. S286/T287/S288 phosphorylation is a key regulator of M2 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the 5-Hydroxytryptamine Receptor 4 (M2) were either not exposed or exposed to 100 µM carbachol for 30 minutes. Cells were lysed and immunoblotted with the anti-pS286/pT287/pS288-M2 antibody (7TM0014C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-M2 antibody to confirm equal loading of the gel.
