Serine359/Threonine361 is a major phosphorylation site of the M3 Muscarinic Acetylcholine Receptor. The pS359/pT361-M3 antibody detects phosphorylation in response to high-efficacy agonists but not after PKC activation. S359/T361 phosphorylation is a key regulator of M3 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the M3 Muscarinic Acetylcholine Receptor (M3) were either not exposed or exposed to 100 µM Acetylcholine for 30 minutes. Cells were lysed and immunoblotted with the anti-pS359/pT361-M3 antibody (7TM0015B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with an phosphorylation-independent anti-5-HT4 antibody to confirm equal loading of the gel.
