Serine408/Serine409 (S408/S409) is a major phosphorylation site of the VPAC2 receptor. The pS408/pS409-VPAC2 antibody detects phosphorylation in response to high-efficacy agonists. S408/S409 phosphorylation is a key regulator of VPAC2 desensitization, β-arrestin recruitment and internalization.

Upper panel, HEK293 cells stably expressing the VPAC2-Receptor (VPAC2) were either not exposed or exposed to 1 µM of specific VPAC-Receptor agonist PACAP-27 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS408/pS409-VPAC2 antibody (7TM0372A) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-VPAC2 antibody (7TM0372N) at a dilution of 1:1000 to confirm equal loading of the gel.

HEK293 cells stably expressing the VPAC2-Receptor (VPAC2) were either not exposed or exposed to 1 µM of specific VPAC-Receptor agonist PACAP-27 and immunocytochemically stained with the anti-pS408/pS409-VPAC2 antibody (7TM0372A) at a dilution of 1:200. Note, Serine408/Serine409-phosphorylated VPAC2 receptors were not detectable in untreated cells (0 min). Serine408/Serine409-phosphorylated VPAC2 receptors were seen at the plasma membrane after 5 min PACAP-27 stimulation.
