Serine435/Threonine438 (S435/T438) is a major phosphorylation site of the VPAC1 receptor. The pS435/pT438-VPAC1 antibody detects phosphorylation in response to high-efficacy agonists. S422/S425 phosphorylation is a key regulator of VPAC1 desensitization, β-arrestin recruitment and internalization. S435/T438 also undergoes constitutive phosphorylation.

Upper panel, HEK293 cells stably expressing the VPAC1-Receptor (VPAC1) were either not exposed or exposed to 1 µM of specific VPAC1-Receptor agonist PACAP-27 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS435/pT438-VPAC1 antibody (7TM0371C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-VPAC1 antibody (7TM0371N) at a dilution of 1:1000 to confirm equal loading of the gel.
