The non-phospho-ACKR3 receptor antibody is directed against the carboxyl-terminal tail of mouse, rat and human ACKR3. It can be used to detect total ACKR3 receptors in Western blots independent of phosphorylation. The non-phospho-ACKR3 antibody can also be used to isolate and enrich ACKR3 receptors from cell and tissue lysates. The ACKR3 antibody can also be used to detect ACKR3 in formalin-fixed, paraffin-embedded tissue sections by immunohistochemistry.

Figure 1. Validation of the Atypical Chemokine Receptor 3 in transfected HEK293 cells. Native HEK293 cells (MOCK) or HEK293 cells stably expressing the Atypical Chemokine Receptor 3 (ACKR3) were lysed and immunoblotted with the phosphorylation-independent c-terminal anti-ACKR3 antibody (7TM0080N-GP) at a dilution of 1:1000.

Sections were dewaxed, microwaved in citric acid, and incubated with anti-ACKR3 (Atypical Chemokine Receptor 3) antibody (7TM0080N-GP) at a dilution of 1:100. Sections were then sequentially treated with biotinylated anti-guinea pig IgG and avidin-biotin solution.Color was developed by incubation in 3-amino-9-ethylcarbazole (AEC), and sections were counterstained with hematoxylin.

Sections were then incubated with anti-ACKR3 (non-phospho-Atypical Chemokine Receptor 3) antibody (7TM0080N-GP) at a dilution of 1:100 and sequentially treated with biotinylated anti-rabbit IgG and avidin-biotin solution. Sections were then developed in 3,3-diaminobenzidine (DAB)-glucose oxidase and lightly counterstained with hematoxylin. Note, only in sections without peptide incubation ACKR3 receptors were uniformly detected at the plasma membrane of cells.
