Threonine348 (T348) is a major phosphorylation site of the somatostatin receptor 3 (SST3). The pT348-SST3 antibody detects phosphorylation in response to high- and low-efficacy agonists but not after PKC activation. T348 phosphorylation is a key regulator of SST3 desensitization, β-arrestin recruitment and internalization.

Figure 1. Agonist-induced Threonine348 phosphorylation of the Somatostatin Receptor 3. Upper panel, HEK293 cells stably expressing the Somatostatin Receptor 3 (SST3) were either not exposed or exposed to 1 μM SRIF (somatotropin-release inhibiting factor) or 1 μM of artificial somatostatin receptor agonist Octreotide for 30 minutes. Cells were lysed and immunoblotted with the anti-pT348-SST3 antibody (7TM0357B) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-SST3 antibody (7TM0357N-WB) at a dilution of 1:1000 to confirm equal loading of the gel. Note, SST3 receptor forms stable dimers.
